Description:

ZINC12427628 (Compound '628) is a potent, selective positive allosteric modulator of M2 mAChR with EC50 of 1.1 uM; enhances the binding of N-methyl scopolamine (NMS), while sparing the endogenous agonist acetylcholine, slows the dissociation rate of NMS from the M2 mAChR by 50-fold; the selective allostery between '628 and NMS was retained in membranes from adult rat hypothalamus and in neonatal rat cardiomyocytes.
